exosome的小鼠实验 我又要重新开始了
本帖最后由 hzangs 于 2015-7-12 19:01 编辑经过了接近3个月的休整和方案调整。我又要开始跳坑了。我要开始做exosome training 小鼠微环境的动物实验了。上次一口气做了近3个月，每个周分离二百多微克的exosome，这次又要来了。过来立个帖子，激励一下自己!!!
PS:书于2015-7-12 小鼠实验已经结束，通过膝盖处穿刺分别注射高、低转移细胞株分泌的exosome， training小鼠骨微环境，每周2次，一次3ug/只，持续一个月。之后心脏注射中等转移能力肿瘤细胞株构建骨转移模型。追踪转移情况，持续6周。
结果，注射高转移细胞株exosome的一组小鼠转移能力确实比另一组要高一些。但是BLI检测转移信号强度差异在2倍左右，差异倍数过小。因此动物实验提前结束，没有进行后续的继续研究。准备从体外实验入手开始研究exosome是否对转移具有影响。
建议大家以后可以考虑先从体外实验入手，再逐渐上动物实验，可能会更靠谱些。
Based on the assumption that 100 μg of T-exosomes are released to the peripheral tissues continuously, we used 100 μg of exosomes for injection experiments. T-exosomes (100 μg) isolated from TS/A or 4T-1 tumor removed at day 21 post tumor cell injection were injected into BALB/c mice via the tail vein twice weekly for three weeks.请问这个意思是说三周共注射100ug的外泌体吗？. Your exosome should be labeled using fluorescence or luciferase or others. Otherwise, you will not know how much you dosage into mouse? By the way, protein alone (BAC) is not a reliable method to quantify exosome, specially if your exosome purity is not too high. Labeled exosome (engineering exosome) will help you to get ride of this issue and figure out where the dosage exosome is homing, in liver or tumor or spleen? In addition, how do you dosage exosome? hydrodynamic IV, IP and oral. the routine is issue for dosage. The cell type for exosome production is another issue too. In short, cell type ( immune cell or solid tumor cell), the dosage routine, and labeling exosome should be considerated.

我是用蛋白定量的方法来衡量exosomes的量。这次试验基本上每周收100-200ug exosomes。
我是做肿瘤细胞exosomes的，基本上需要15cm dish 20盘-25盘 做小鼠？是在体exosome实验吗？那你们有没有从组织中试图提取exosome啊？还是提取exosome回注体内的方法？ 向荣y 发表于 2015-4-15 17:45

0.2 ug/ul 也不算低了，100ul 也有20ug了。
我打老鼠是从尾静脉带进去，每只在2-3ug 支持！有趣有又有难度的实验！ 怒赞！ 很弱弱地问，啥是 exosome training 小鼠微环境？

哦哦，我用的25cm2的培养瓶养的干细胞，但是外体浓度好低啊，160ML的上清浓缩到100微升PBS-exosomes，BCA测的浓度才0.2微克每微升，作用到细胞上完全看不出变化。所以想问一下你的外体浓度是多少呢？要做动物实验，外题的量是不是要更大一些？
