The different cells will produce the different amount of exosome. The average amount of exosome in cell culture supernatant is around 10^6p/ml. I do not know which kit you use now. If you use PEG-based precipitation kit, you got some, but lose almost half of them. It is good idea to start a large of volume at this point. Labeling exosome and then spike-in will help you figure out the efficiency of isolation. Checking protein/miRNA and counting particle will help.
