【投稿经验】审稿人对SBI试剂盒的疑问及解决

本人正在投一篇Scientific Reports（水刊，求大神不要嘲笑）
用SBI试剂盒提取的exo，审稿人针对这个方法提出了如下comments，分享给各位群里的战友：

In Fig. 1, western blot analysis was performed using cell lysates and exosome lysates as starting materials. However, purified exosomes, especially if a polymer-based extraction kit such as the Exo-Quick kit is used, are heavily contaminated with carry-over proteins and cell debris from the initial DC culture media. Therefore, the authors should add a control were the same exosome samples are first digested with proteinase K to eliminate extra-exosomal contaminants and then lysed and used for western blot analysis. This control would help determine whether the indicated markers are of exosomal origin or not.  




补实验中。。。都毕业了还要给自己擦屁股，唉。。。

值得借鉴的意见。

想问一下这里面用的proteinase K 会不会也降解了exosome 膜上的蛋白呢，还有这个降解时间怎么控制呢。

我也不知道咋控制时间，打算查查资料先试试。。。

这个方法我在文献里也没看到过。
但考虑到我现在是补实验，肯定要尊重审稿人的意见了，打算先这样做起来。
这个审稿人既然提出了这个建议，或许也是外泌体的“业内人士”。。。

对啊，起码没有全盘否定试剂盒的提取效果。
