关于原代细胞提取外泌体？

我们课题组研究乳腺癌细胞的外泌体，近期老板想研究原代细胞的外泌体。原代细胞是从病人离体的组织分离出来的？想问有没有谁做过这个提取呢？

Our lab routinely collect exosomes/Evs from primary rat astrocytes. Using serum-free medium (SFM), incubating for 24 hours, and collecting by serial centrifugation/ultracentrifugation. But some people argued that it's better to use medium with EV-free serum rather than SFM.

Thank you very much. As what you respond, we consider that if we use SFM for primary cells, wthere the cells could survival? 24h or less? Later, I will try to compare SFM and EV-free serum.
Additionally, I want to collect the EVs through Life KIT, which did you think?
And, how much  cell supernatant with how many cells did you use for collecting?
